Unlike pregnant adult women, pregnant adolescent girls cannot maintain glycine flux during late pregnancy because of decreased synthesis from serine

During pregnancy, glycine and serine become more important because they are the primary suppliers of methyl groups for the synthesis of fetal DNA, and more glycine is required for fetal collagen synthesis as pregnancy progresses. In an earlier study, we reported that glycine flux decreased by 39 % from the first to the third trimester in pregnant adolescent girls. As serine is a primary precursor for glycine synthesis, the objective of this study was to measure and compare glycine and serine fluxes and inter-conversions in pregnant adolescent girls and adult women in the first and third trimesters. Measurements were made after an overnight fast by continuous intravenous infusions of 2H2-glycine and 15N-serine in eleven adolescent girls (17·4 (se 0·1) years of age) and in ten adult women (25·8 (se 0·5) years of age) for 4 h. Adolescent girls had significantly slower glycine flux and they made less glycine from serine in the third (P<0·05) than in the first trimester. Baby birth length was significantly shorter of adolescent girls (P=0·04) and was significantly associated with third trimester glycine flux. These findings suggest that the pregnant adolescent cannot maintain glycine flux in late pregnancy compared with early pregnancy because of decreased synthesis from serine. It is possible that the inability to maintain glycine synthesis makes her fetus vulnerable to impaired cartilage synthesis, and thus linear growth.

Adaptation of in vivo amino acid kinetics facilitates increased amino acid availability for fetal growth in adolescent and adult pregnancies alike

During pregnancy, adult women with a normal BMI synthesise extra amino acids after an overnight fast by increasing body protein breakdown and decreasing amino acid oxidation. It is not known whether adolescent girls can make these adaptations during pregnancy. The present study aimed to measure and compare the protein, glutamine and alanine kinetics of adult women and adolescent girls at early-, mid- and late-pregnancy. Kinetics were measured in the overnight fasted state using intravenous infusions of 13C-leucine, 15N-glutamine and 15N-alanine in ten adults and twenty adolescents aged 14–17 years in the first and second trimesters (phase 1 study) and infusions of 13C-leucine and 15N2-urea in ten adults and eleven adolescents aged 16–17 years in the first and third trimesters (phase 2 study). In phase 1 study, there were no significant differences between the groups with regard to any of the kinetic parameters measured. In both groups, leucine flux increased (P< 0·05), the percentage of leucine flux oxidised decreased (P< 0·05) and non-oxidative leucine disposal to protein synthesis increased (P< 0·05) from the first to the second trimester. In phase2 study, leucine flux was significantly slower (P< 0·05) in the adult group than in the adolescent group during both trimesters, and whole-body leucine flux and non-oxidative leucine disposal increased significantly in the adolescent group (P< 0·05, respectively) and were higher in the adult group from the first to the third trimester. These results suggest that similar to their adult counterparts after an overnight fast, adolescent girls with a normal BMI provide extra amino acids required for net protein deposition during pregnancy by increasing protein breakdown and decreasing amino acid oxidation.

Introduction
Metalliferous soils provide very restrictive habitats for plants due to phytotoxicity, resulting in severe selection pressures. Species comprising heavy-metal plant communities are genetically altered ecotypes with specific tolerances to, e.g., cadmium, copper, lead, nickel, zinc and arsenic, adapted through microevolutionary processes. Evolution of metal tolerance takes place at each specific site (Ernst 2006). A high degree of metal tolerance depends on the bioavailable fraction of the metal(loids) in the soil and the type of mineralization. At extremely high soil metal concentrations, especially on polymetallic soils, even metal-tolerant genotypes are not able to evolve extreme tolerances to several heavy metals simultaneously. Adapted genotypes are the result of the Darwinian natural selection of metal-tolerant individuals selected from surrounding non-metalliferous populations (Antonovics et al. 1971; Baker 1987; Ernst 2006). Such selection can lead ultimately to speciation and the evolution of endemic taxa. Heavy-metal tolerance was first reported by Prat (1934) in Silene dioica and demonstrated experimentally in grasses by Bradshaw and co-workers in Agrostis spp. and by Wilkins in Festuca ovina in the late 1950s and 1960s (see Antonovics et al. 1971) and from the early 1950s onwards in the herb Silene vulgaris by Baumeister and co-workers (see Ernst 1974). Metal-tolerant plants avoid intoxication by an excess of heavy metals by means of special cellular mechanisms, as long as the soil metal levels do not exceed the levels of metal tolerance (Ernst 1974; Ernst et al. 2004). They can thus thrive on soils that are too toxic for non-adapted species and ecotypes.
Determining uptake of ‘non-labile’ soil cadmium by Thlaspi caerulescens using isotopic dilution techniques

We assessed the ability of several populations of the metal-hyperaccumulator species, Thlaspi caerulescens, to mobilize non-labile cadmium in soils historically contaminated by Pb/Zn mine spoil or sewage sludge. Radio- labile Cd was determined chemically as an ‘E-value’, [CdE], and biologically as an ‘L-value’, [CdL]. For comparison, chloride-extractable Cd, [Cdchlor], was also determined using 1 M CaCl2 as a single-step soil extractant. Values of [CdL] were measured for six populations of T. caerulescens that varied substantially in their ability to assimilate soil Cd, and a non-accumulator species with a similar growth habit, Lepidium heterophyllum. Seeds were sown in soil spiked with 109Cd and grown for 9–12 wk in a controlled environment room. Values of [CdL] were determined from the specific activity of 109Cd and concentration of Cd in the plant leaves. For the six soils studied, [CdE] ranged from 4.9 to 49% of total soil Cd [CdT]. Values of [CdL] were, in general, in close agreement with both [CdE] and [Cdchlor] and substantially less than [CdT]. However, [CdL] showed no correlation with the concentration of Cd in plant tissue, [Cdshoot]. This suggests that, in the soils studied, T. caerulescens did not mobilize non-labile soil Cd by producing root exudates or altering rhizosphere pH. The results imply that there may be significant restrictions to metal bioavailability, even to hyperaccumulator species, in heavily contaminated soils in which a large proportion of the metal may be present in ‘non-labile’ forms.

Seedling mortality of Alyssum serpyllifolium ssp. lusitanicum and A. murale, both nickel hyperaccumulators, was reduced by increasing concentrations of metal within plant tissues when inoculated with the fungi Pythium mamillatum or P. ultimum, both of which cause damping-off disease of seedlings. Pythium mamillatum, isolated from nickel-rich serpentine soil, was more tolerant of nickel than P. ultimum, isolated from low-metal control soil, and was more pathogenic than P. ultimum towards seedlings containing high concentrations of metal. These results support the hypothesis that metal hyperaccumulation by plants is closely linked to increased protection against disease.
Positive responses to Zn and Cd by roots of the Zn and Cd hyperaccumulator Thlaspi caerulescens

The effects of localized zinc (ZnO) and cadmium (CdS) enrichment on the allocation of root biomass, root length and partitioning of current assimilate within root systems of the Zn hyperaccumulator Thlaspi caerulescens were investigated using a rhizobox system. The rhizoboxes contained either homogeneous soil or juxtaposed control and metal-enriched soil. In the heterogeneous treatments the Zn-enriched soil contained 250, 500 or 1000 mg Zn kg−1. The plants consistently allocated c. 70% of their total root biomass and length into the metal-enriched soil. Moreover, 70% of the current assimilate (14C) allocated to the roots was localized in the Zn-enriched soil of the heterogeneous treatments. The concentration of Zn (250–1000 mg kg−1) in the enriched soil had no effect on these patterns of root allocation, nor were there significant effects of the Zn treatments on the total root or shoot biomass of the plants. The positive responses to the localized metal treatments were therefore characterized by a concomitant reduction in root allocation into the control soil. In contrast to T. caerulescens, when grown with juxtaposed control and Zn-enriched soil the non-accumulator Thlaspi arvense showed reduced root allocation into a patch enriched with 500 mg kg−1 Zn. In a further experiment, two populations of T. caerulescens that differ in their abilities to accumulate Cd were grown with juxtaposed control and Cd-enriched soil. The plants from a population that accumulated Cd also showed increased root biomass and root length allocation into the Cd-enriched soil. Plants from the population that did not accumulate Cd showed no such increase. The possibility that T. caerulescens forages for metals, and the precision of its root allocation with respect to localized metal enrichment is highlighted. The significance of these findings for the selection of hyperaccumulator plants for use in the phytoextraction of Zn and Cd from mine spoils (phytomining) and the phytoremediation of heterogeneous contaminated soils are discussed.

Plants known as hyperaccumulators take up and sequester high concentrations of potentially toxic elements from metalliferous soils. We tested the hypothesis that zinc hyperaccumulation benefits plants by deterring herbivory. In laboratory feeding trials, three species of herbivores were allowed to choose between Thlaspi caerulescens (Brassicaceae) plants grown in low-Zn and Zn-amended culture solution. Locusts (Schistocerca gregaria), slugs (Deroceras caruanae), and caterpillars (Pieris brassicae) all showed significant preferences for plants with lower foliar Zn concentrations. Such differential feeding could result in selection pressures favouring the evolution of hyperaccumulation. The findings are also relevant to current proposals to exploit hyperaccumulation as a means of remediating metal-contaminated soils.
